En su primer viaje, Colón reclamó a San Salvador, Cuba e Hispaniola como posesiones españolas. Construyó un fuerte y dejó atrás a los soldados españoles para buscar oro en La Española, mientras regresaba a España. (Estos hombres fueron asesinados más tarde por los habitantes de la isla por maltratarlos). En su segundo viaje, Colón llevó a un millar de colonos españoles a establecerse en La Española. Esta fue la primera colonia europea en el "Nuevo Mundo". Estos colonos lucharon entre sí y con los habitantes de la isla. Fueron codiciosos y se quejaron de que no había suficiente oro para hacerlos ricos a todos. Se les dio tierra y se les permitió obligar a los indígenas a trabajar para ellos, pero todavía no estaban satisfechos. Los colonos también fueron responsables de la introducción de enfermedades epidémicas extranjeras, como la influenza, la viruela, el sarampión y el tifus, que redujeron drásticamente la población indígena en el Caribe dentro de los 50 años.
